Position: Scientist 1/2 (contractor)
Department: Structural Biology
Summary:
The Department of Structural Biology within Genentech Research and Early Development (gRED) has an opportunity for a highly motivated Scientist 1 or 2(contract) with expertise in protein biochemistry and structural biology to join our collaborative group. The Scientist’s work will be focused on generation and optimization of recombinant proteins for drug discovery and research. Hands-on experience with protein crystallization and/or sample preparation for cryoEM is highly desired although not critical. The successful candidate will be responsible for generating high-quality protein reagents to support small and large molecule drug discovery, high throughput screening, assay development and structural biology, working with multiple groups across Genentech. This role will leverage considerable bench work combined with the use of robotic platforms for protein purification, crystallization and protein characterization by biophysical & biochemical methods. The individual will work independently at the bench and collaborate with colleagues across various functional areas to generate and communicate data to further research and drug discovery.
Who we are:
The Genentech Structural Biology Department integrates macromolecular crystallography, CryoEM and NMR spectroscopy to drive drug discovery and uncover fundamental biology. Our department offers a highly interdisciplinary environment with unparalleled resources and unique opportunities for scientific collaboration and growth. We explore the structural and functional properties of proteins of therapeutic interest, and their interactions with ligands, substrates, and binding partners. Our group is involved in every step from construct design and protein purification to structure determination, in-depth analyses and drug design. We collaborate with scientists throughout the Research organization to advance both traditional “small molecule” and protein-based drug discovery as well as to pursue basic research questions regarding structure and function.
Required qualifications:
- BS/MS degree with 3-5 years of experience or a PhD with 0-2 years of experience in Biochemistry, Chemical Biology, Structural Biology or a related discipline.
- Hands-on experience designing and optimizing the purification of challenging protein targets for structural and/or functional studies.
- Demonstrated expertise with modern protein purification and chromatography techniques, and knowledgeable with operating AKTA chromatography systems as well as analytical HPLC systems.
- Relevant experience in purifying challenging proteins or with purifying a wide range of protein families.
Excellent communication, interpersonal and organizational skills.
- Team-oriented, collaborative nature with fearless attitude to tackle long standing difficult problems in research and drug discovery.
Additional desired qualifications:
- Strong candidates will have many of the following skills and attributes:
- Experience in protein expression systems such as E coli, mammalian or insect.
- Experience in purification and assembly of multi-protein complexes.
- Hands-on experience in protein crystallization, crystal optimization and harvesting for data collection.
- Experience in cryoEM sample and grid preparation.
- Experience with methods to characterize protein quality and purity, such as mass spectrometry, SEC-MALS/DLS, endotoxin assays, and DSF/thermal shift assays.
- Experience with biophysical and biochemical assays.
